ABSTRACT

Objective:To optimize the extraction parameters for anthraquinones in Semen Cassiae by Box-Behnken experimental design. Methods:Based on single factor screening, a three-factor and three-level Box-Behnken experimental design was employed with the ethanol concentration, material to liquid ratio and extraction time as the independent variables. The dependent variables including the extration rates of chrysophand and aurantio-obtusin were transformed into desirability mathematically by Hassan 's method. The mathematical relationship was established between the dependent variables and the independent variables. The optimum experimental conditions were selected from the stationary point of the response surfaces. Results:The optimum extraction conditions were as follows:12-fold amount of 44% ethanol as the solvent, extracted three times with 2. 65h for each time. The extraction rate of chrysophanol and aurantio-obtusin was 1. 921% and 3. 244%, respectively. Conclusion:The measured value is close to the predicted one,which indi-cates the comprehensive extraction parameters optimized by Box-Behnken experimental design can be used for the extraction of chry-sophanol and aurantio-obtusin from Semen Cassiae.

ABSTRACT

A quality control method of Semen Cassiae was established by using naphthopyrone reference extract(NRE). Meanwhile, the feasibility about NRE replacing single component reference in quality control of traditional Chinese medicine was explored. After NRE of Semen Cassiae being prepared by chromatographic separation technology, we determined the three main components, cassiaside B2, rubrofusarin-6-O-β-D-gentiobioside and cassiaside C. In the meantime, an HPLC analytical method, based on the NRE as standard substance, was developed to determinate the contents of three main components in Semen Cassiae. T-test was used for comparison of the determination results of the two methods(single chemical component and NRE as reference substances, respectively), and the T-test result demonstrated that there was no significant difference between the two methods. The results developed scientific basis for the application of NRE of Semen Cassia in the quality control, which could be applied for the quality control of traditional Chinese medicine using reference extract substituting single chemical reference, and provide a new research model for the quality control of Chinese medicine.

ABSTRACT

Objective Obese patients with type 2 diabetes mellitus ( T2DM) characterized by hyperglycemia are li-able to more severe myocardial infarction .Semen Cassiae is proved to reduce serum lipid level .This study was to investigate whether the Semen Cassiae extract (SCE) reduces myocardial ischemia and reperfusion (MI/R) injury with or without diabetes and the underlying mechanisms .Methods The high-fat diet-fed streptozotocin ( HFD-STZ) rat model was used as T2DM model.Normal and DM rats received SCE treatment orally (10 mg/kg/day) for 1 week.Subsequently these animals were subjected to MI /R.Results Compared with the normal animals , DM rats showed increased plasma total cholesterol ( TC) and triacylglycerol ( TG) , more severe MI/R injury and cardi-ac functional impairment .SCE treatment significantly reduced the plasma TC and TG , improved the instantaneous first derivation of left ventricle pressure and reduced infarct size , decreased plasma creatine kinase and lactate dehydrogenase levels, and reduced apoptosis index at the end of reperfusion in diabetic rats .Moreover, SCE treat-ment increased the antiapoptotic protein Akt and stimulated ERK 1/2 phosphorylation .Pretreatment with a PI3 K in-hibitor wortmannin or an ERK1/2 inhibitor PD98059 not only blocked Akt and ERK1/2 phosphorylation, but also inhibited the cardioprotective effects of SCE .However , SCE treatment did not show any effects on the MI/R injury in the normal rats.Conclusions SCE effectively improves myocardial function and reduces MI/R-induced injury in diabetic but not normal animals , which is potentially attributable to the reduced TC/TG levels and the triggered cell survival signaling Akt and ERK 1/2 .

ABSTRACT

Objective To observe the clinical curative effect of Semen Cassiae application on navel in treating essential hypertension. Methods Totally 1674 cases of hypertension, according to visiting sequence, were randomly divided into treatment group (864 cases) and control group (810 cases). The control group was given hydrochloride enalapril 10 mg orally, 2 times a day. The treatment group was treated with Semen Cassiae application on navel, replaced per 24 h. The two groups were treated with moderate exercise, reasonable diet, and the course was 6 months. The clinical symptoms, blood pressure, blood lipid and blood rheology of the two groups were measured before and after treatment. Results To compare the symptoms efficacy, total effectiveness of treatment group was 95.14%(822/864), the control group was 71.11%(576/810), there was a significant difference (P<0.01). To compare antihypertensive effects, the total effective rate of the treatment group was 95.60% (826/864), the control group was 73.46% (595/810), there was a significant difference (P<0.01). The change of blood pressure in the treatment group was significantly better than the control group (P<0.01). TC, TG, HDL-C, LDL-C and blood rheology of the treatment group was significant different between before and after treatment (P<0.01). Conclusion On the basis of western medicine, Semen Cassiae application on navel played a double effect of acupoint and drug. It is more effective in lowering blood pressure, improving the clinical symptoms, blood lipid and blood rheology indexes.

AIM@#To establish a more comprehensive and suitable procedure for the quality control of Semen Cassiae which can be used to supplement the evaluation procedure adopted by the Pharmacopoeia of the Peoples Republic of China.@*METHODS@#A HPLC assay-based comprehensive quality evaluation procedure for Semen Cassiae using three bioactive compounds including anthraquinones and naphthopyrones, i.e., chrysophanol-1-O-β-D-glucopyranosyl-(1→3)-β-D-glucopyranosyl-(1→6)-β-D-glucopyranoside (1), rubrofusarin-6-O-β-D-gentiobioside (2) and toralactone-9-O-β-D-gentiobioside (3) as the index components was established. The resultant data were further analyzed by principal component analysis (PCA) and data distribution methods using software SPSS 16.0.@*RESULTS@#Sixty-six batches of Semen Cassiae obtained from various regions of China were analyzed with the procedure. Based on the test results of these batches, the content limits of the three bioactive compounds in Semen Cassiae were proposed.@*CONCLUSION@#The procedure established herein is more comprehensive and appropriate for the quality evaluation of Semen Cassiae commercially available in China, and can be used as a useful supplement to the current official method in China for the quality evaluation of Semen Cassiae.

ABSTRACT

Objective To explore the determination of elements selenium content in Se Semen Cassiae. Method Selenium content in Se Semen Cassiae was measured by graphite atomic absorption spectrophotometer. Result and Conclusion An appropriate extraction method and digesting condition were screened out. The determined results were accurate when the digested sample was dissolved and metered volume with 1% HNO3 which containing 0.1% Tween 80.

ABSTRACT

OBJECTIVE:To investigate the effects of different concentration of methanol on 5 anthraquinone constituents in Semen Cassiae.METHODS:Semen Cassiae was extracted with different concentration of methanol(95%,80% and 30%).The contents of aloe-emodin,rhein,emodin,chrysophanol and physcione in Semen Cassiae were determined by HPLC.RESULTS:The contents of aloe-emodin,rhein,emodin,chrysophanol and physcione were 0.001 0 ~0.018 1 mg?g-1,0.010 4~0.063 7 mg?g-1,0.020 9~0.253 5 mg?g-1,0.003 1~0.915 7 mg?g-1 and 0.005 1~0.126 3 mg?g-1,respectively,The average recoveries were 96.34%~97.16% and the RSDs of all were less than 3%(n=6).CONCLUSION:There were great differences in the contents of the 5 anthraquinone constituents in Semen Cassiae extracted with different concentration of methanol,and a best extraction efficacy was achieved for Semen Cassiae extracted with 95% methanol while the worst extraction efficacy was noted if extracted with 30% methanol.

Objective To establish a method for the quality control of Semen Cassiae.Methods A high performance liquid chromatographic method was developed to establish the fingerprint of Semen Cassiae,and 32 samples from various batches of Semen Cassiae were analyzed.Cluster analysis and principal component analysis were applied to study HPLC fingerprint and chemical pattern recognition.Results The fingerprint of Semen Cassiae was set up.Conclusion The method could be used for the quality control and comprehensive evaluation of Semen Cassiae.

ABSTRACT

Objective To evaluate the quality of Semen Cassiae from different habitats objectively. Methods To determine the content of chryso-phanic according to ChP and establish HPLC fingerprints with the gradient elution solvent composed of acetonitrile and 1% HAC. A C18 column (250 mm?4.6 mm, 5 ?m) was used, flow rate: 1 mL/min, detecting wavelength: 254 nm, and the column temperature: room temperature. The clustering analysis was carried out by SAS software according to the content of chrysophanic and similarity of HPLC fingerprints obtained by the software of similarity analysis. Results The established HPLC fingerprint has desirable precision, reproducibility, and stability. The content of chrysophanic and HPLC fingerprints of Semen Cassiae from various habitats are different, which differs from the habitats. The content range of chrysophanic in Semen Cassiae is 0.037%-0.170% and the similarity is 0.864-0.962. Conclusion The method indicates the difference of the chemical component in Semen Cassiae from various habitats and can be used as a quality control method for Semen Cassiae.

MethodsChromatographic fingerprint of Semen Cassiae was investigated by RP-HPLC and the gradient elution mode was applied to chromatographic separation. Data were analysed by fingerprint similarity evaluation software to compare the similarity of the samples. ResultsThe HPLC fingerprint of Semen Cassiae was established preliminarily. ConclusionHPLC Fingerprint method is repeatable and can be used in quality control of Semen Cassiae.

ABSTRACT

Objective To investigate the pharmacological actions related with therapeutic effect and indications of crude and processed semen cassiae,and to compare the differences between them.Methods Serum contents of ALT and AST were determined in acute liver injury mice induced by CCl4,the intestinal propulsion of Evans blue was measured in normal mice and in constipation mice,and stool properties,defecation latency and defecate amounts in constipation mice were also investigated.Results Both crude and processed semen cassiae showed obvious liver-protective effects,and decreased the contents of ALT and AST in serum,but processed semen cassiae was better than crude in protecting liver.Both of them increased the intestinal propulsion in normal and constipation mice,improved the stool properties,shortened the defecate latency and increased the defecate amounts in constipation mice.Conclusion Both crude and processed semen cassiae have the effects of protecting liver,lubricating bowel and promoting defecation.The processed is superior to the crude in liver protection and enzyme decrease,and they have similar effect in lubricating bowel and promoting defecation.

ABSTRACT

AIM: To evaluate the characteristic and explore the mechanism of MAE on Semen Cassiae by comparing MAE with commonly used extraction method. METHODS : The amount of anthraquinone was determined by spectrophotometer. The surface and cross section of Semen Cassiae were observed by microphotography. RESULTS : Among the four methods,the efficiency of MAE is 16 times that of ultrasonic extraction,3 times that of Sohlex extraction and 1.1 times of decocting by water,respectively. Micrographs taken after extraction differed markedly indicated that the degree of damage varied considerably. CONCLUSION : The MAE method is more advantageous than other traditional extraction methods (Soxhlet extraction and ultrasonic extraction) with regard to the extraction yield and the time and cost of the procedure.

ABSTRACT

OBJECTIVE:To study the HPLC fingerprint profiles of crude and processed Semen Cassiae so as to provide basis for the establishment of chromatographic fingerprint of Semen Cassiae.METHODS: The samples of Semen Cassiae were hydrolyzed in the 1.5 mol?L-1 hydrochloride acid and then reflux extracted with chloroform.The HPLC analysis was run in gradient elution.The whole information of single-wavelength chromatographic fingerprint,two-dimension information data at full wavelength 220 nm~550 nm and the data of 18 fingerprint peaks compared with chrysophanol were treated by cluster analysis.RESULTS:It was observed that the contents of several chemical components were higher in the crude or processed Semen Cassiae than in chrysophanol.There were differences between the crude and the processed Semen Cassiae in HPLC fingerprint profiles though they were of good similarity.CONCLUSION: The HPLC fingerprints of liposoluble constituents were obtained with good resolution under the experimental chromatographic system.Three pure chromatographic peaks of chrysophanol,emodin and physcion have been identified.More reliable cluster result can be obtained from two-dimension information data at full wavelength 220 nm~550 nm than from the whole information of single-wavelength chromatographic fingerprint or from the information of chromatographic fingerprint of characteristic peaks.
